It can be about an astronaut leaving to space, in a clear alussion to the moon landing. Or it can be that Major Tom is actually a junkee getting a fix and, possibliy, an overodse. David Bowie has always been ambiguous about this, but in his 1980 song “Ashes To Ashes” Bowie sings “we know Major Tom’s a junkee”, which leads me toward the second interpretation.
The name of the song is an obvious reference to the film 2001: A Space Odyssey.
